General description

Tyrosine-protein phosphatase non-receptor type 11 (EC 3.1.3.48; UniProt P35235; also known as Protein-tyrosine phosphatase SYP; SH-PTP2, SH2 domain-containing protein tyrosine phosphatase-2, SHP-2, Shp2) is encoded by the Ptpn11 (also known as Syp) gene (Gene ID 19247) in murine species. SHP-2/SHPTP-2 is a non-receptor protein tyrosine phosphatase involved in a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. SHP-2/SHPTP-2 contains two tandem Src homology-2 (SH2) domains that mediate substrates interaction, followed by a protein tyrosine phosphatase PTP domain. Peptide library-based study reveals similar substrate specificities of SHP-2 and the related SHP-1. Both enzymes exhibit preference for acidic and hydrophobic residues flanking the N- and C-terminal sides of target tyrosine residue. Mutations in PTPN11 gene are known to cause Noonan syndrome and acute myeloid leukemia in human.
